Methods for the Enrichment and Concentration of Waterborne Viruses
Abstract
This standard applies to the enrichment and concentration of bacteriophages, norovirus, coxsackievirus, rotavirus, enterovirus, adenovirus, SARS-CoV-2 and other viruses in source water, drinking water, recreational landscape water and wastewater. Testing laboratories shall conduct prefiltration according to sample turbidity, add magnesium chloride and adjust pH, then use anion-exchange membrane adsorption, TGBE elution and ultrafiltration concentration. Process control viruses and nucleic acid amplification controls shall be used for quality control, followed by viral nucleic acid extraction, purification and RT-qPCR testing. RNase-free sterile consumables shall be used to prevent cross-contamination, and laboratory biosafety requirements shall be met.
